多个已知全同胞参与的全同胞关系排除方法

摘要:

目的建立一种多个已知全同胞参与的全同关系排除方法—第五等位基因排除法。方法本方法原理:当2个以上全同胞参与鉴定时,若已知全同胞和被鉴定人在某一个STR位点出现了多于5个的等位基因(含5个),则该位点不符合遗传规律,倾向于排除被鉴定人;如果检出3个以上这类位点则排除全同胞关系。为了评估该方法的检验效能,本研究用GoldeneyeTM20A中19个STR位点对日常检案中已明确13组2-全同胞和5组3-全同胞的体系进行检验,与100个无关个体进行比对并统计其排除率。结果用19个STR位点检测13组2-全同胞与100个无关个体的排除率为47.77%,5组3-全同胞的排除率为88.00%。若已知全同胞在某一基因座提供3个以上等位基因则称之为有效STR,有效STR数目增多时排除率也增加。结论第五等位基因排除法结论明确,可推广于多人参与的全同胞鉴定。

  • Objective To establish an approach for exclusion of full sibling relationship concerning the participation of multiple known full-siblings —— the 5thallele exclusion method. Methods The principle of this method is: when more than 2 known full-siblings joined in the examination for the full-sibling relationship identification of a suspect,the total number of different alleles from all the tested individuals should be less than 5 at one STR locus. Otherwise,it does not conform to the Mendel genetic law. If this condition is detected in three or more STR loci,then the suspect should be excluded,i. e. the suspect was not a fullsibling of the known-siblings. In addition,the exclusion power of this method was evaluated by analyzing 13 groups of 2 known full siblings and 5 groups of 3 known full siblings with the GoldeneyeTM20 A kit after comparison with 100 unrelated individuals. Results When 2 known full siblings participated in the examination,the exclusion rate was 47. 77%,and the exclusion rate rose to 88. 00% when 3 known full siblings participated in. The locus was named as effective STR when 3 or more alleles were offered by the known full siblings. The number of effective STR increased,the exclusion rate increased. Conclusion The 5thallele exclusion method is simple and clear,and this approach could be used in the full sibling identification when2 or more known full siblings participate in the examination.
